Print Runs

Can you discuss print runs? Do pre-sales help? When are they determined? I have a debut novel coming out and I don't know much about this topic.
When I was an agent I was always a bit removed from these discussions, but these two posts from Eric from Pimp My Novel explain a lot about what goes into publishers' initial guess at print run, then what happens at the major accounts, which will prompt publishers to adjust the print run.
