Who's the best person to be an alpha reader?

My WIP should be finished the first re-write in a few weeks and I'd love to hand it off to an alpha reader at that stage. What kind of qualities does a good alpha reader have? I'm guessing loves to read as a start, but what else is important?

US or UK spelling in a query/manuscript

I'm an Australian and I spell using Australian english (customise that colour, would you). If I was submitting to a US agent should the query letter/sample pages/manuscript be re done with US spelling, or is UK/Australian spelling fine?

Should I just look for an agent in the US/UK?

When the time comes, should I be submitting my manuscript to agents in my own country (Australia) or just the US/UK? If I have a local agent, will they be able to get my manuscript published internationally? If so, will they be taking a larger commission for 'foreign' rights?
