I guess it doesn't hurt to have a proposal as well as the completed manuscript (but make it clear that it is completed in the query). I'm probably wrong but I think it depends on the genre. I thought I had a good query letter and when I queried a couple of agents, I put "query: title of my book" in the subject line and within a few minutes had rejections. It may have been my query, I'm slowly learning how to do it right,...
